Natural Hormone FGF19 Unveils New Pathways for Fat Burning

A natural hormone known as FGF19 has demonstrated the ability to enhance fat burning by acting on brain cells, offering a promising avenue for obesity treatments. Researchers have discovered that FGF19 triggers the brain to increase energy expenditure and activate fat-burning cells. This breakthrough could lead to innovative therapies aimed at combating obesity and related metabolic disorders.

Studies conducted on mice reveal that FGF19, produced in the intestine, sends signals to the hypothalamus, a critical brain region for energy regulation. By activating thermogenic adipocytes, or fat cells that expend energy to generate heat, FGF19 helps the body utilize stored fat more effectively. This process not only promotes weight control but also improves blood glucose levels, particularly in obese animals.

Potential for New Obesity and Diabetes Treatments

The findings from this research may pave the way for new medications targeting obesity and diabetes. The objective is to design compounds that mimic the actions of FGF19, similar to how existing drugs like Ozempic utilize natural body substances to regulate appetite. According to the study, FGF19 not only influences appetite but also reduces inflammation and enhances cold tolerance in the body.

However, these benefits are contingent upon the activation of the sympathetic nervous system. When researchers inhibited this system, the positive effects of FGF19 disappeared. Further investigation revealed that exposure to cold temperatures increases the expression of FGF19 receptors in the hypothalamus, suggesting an evolutionary link to temperature regulation.

Professor Helena Cristina de Lima Barbosa from the Obesity and Comorbidities Research Center at the State University of Campinas explains, “FGF19 had already been linked to a reduction in food intake. Our work broke new ground by showing that it also plays an important role by acting on the hypothalamus and stimulating an increase in energy expenditure.”

Addressing the Global Obesity Crisis

The urgency of these findings is underscored by data from the World Atlas of Obesity 2025, which projects that more than 1 billion people worldwide currently live with obesity. If trends continue, this number could exceed 1.5 billion by 2030. Obesity is linked to approximately 1.6 million premature deaths each year from non-communicable diseases. In Brazil, around 31% of the population is classified as obese, highlighting the critical need for effective interventions.

FGF19 primarily originates in the small intestine and plays a significant role in regulating energy metabolism in the liver, influencing bile acid production and the synthesis of glucose and fats. While its effects on the liver are well-documented, the role of FGF19 in the brain has received less attention until now.

The recent study, published in the American Journal of Physiology – Endocrinology and Metabolism, highlights the potential of FGF19 in enhancing energy homeostasis. By stimulating the sympathetic nervous system and increasing thermogenesis in adipose tissue, FGF19 promotes higher energy expenditure.

Lucas Zangerolamo, a doctoral student and first author of the study, noted, “The brain plays an extremely important role in controlling the body’s adiposity. It receives information from peripheral tissues and triggers commands that appear to utilize the sympathetic nervous system to regulate energy expenditure.”

The research team also utilized advanced techniques to analyze public scRNA-seq data, allowing them to identify which brain cells express FGF19 receptors. A key focus moving forward will be to determine how to enhance the body’s natural production of FGF19, as well as to investigate its relationship with inflammation linked to high-fat diets.

As scientists delve deeper into the mechanisms of FGF19 and its receptors, there is hope that these findings will contribute to innovative solutions in the fight against obesity and related health challenges.
